Kcnj4, the mouse ortholog of human KCNJ4, encodes Kir2.3, a strong inward rectifier of the Kir2 family that differs from Kir2.1 and Kir2.2 in having lower single-channel conductance, weaker PIP2 affinity, and pronounced inhibition by intracellular protons, making it the pH-sensitive member of the group. It is enriched in forebrain neurons, particularly in hippocampus and cortex where it contributes to resting conductance and dendritic input resistance, and it is present in atrial myocytes and kidney. Its carboxy-terminal PDZ-binding motif recruits PSD-95 and SAP97, localizing the channel to synaptic and lateral membrane domains. Because Kir2.3 heteromerizes freely with Kir2.1 and Kir2.2 encoded by Kcnj2 and Kcnj12, single-locus nulls are viable and usually show only partial IK1 reduction, so compound conditional alleles or dominant-negative pore knockins are required to unmask function. Align the allele with the endpoint, using forebrain-restricted Cre for excitability studies and pH-sensing residue knockins for gating mechanism.
