Why model Lefty2
Serving as the diffusible inhibitor arm of the left-right circuit, Lefty2 is the mouse ortholog of human LEFTY2 and encodes a secreted, cleaved TGF-beta family protein that captures Nodal and the EGF-CFC coreceptors Cripto and Cryptic, preventing ActRIIB and ALK4 engagement and limiting SMAD2 and SMAD3 output. Because Nodal induces Lefty2 in left lateral plate mesoderm, the pair constitutes a self-regulating activator with a long-range inhibitor that positions the Pitx2 domain and fixes situs. Loss causes embryonic lethality from excess Nodal signaling with expanded mesendoderm, and human variants appear in heterotaxy and complex cardiac malformation. Consequently, conditional and hypomorphic alleles are preferable to germline nulls for laterality work, deletion of the asymmetric enhancer separates Nodal-dependent induction from constitutive expression, and dosage-sensitive knockins probe inhibitor range. Lefty1 is tandemly linked and functionally overlapping, so compound alleles require cis engineering rather than conventional intercrossing.
