Msh6, the mouse ortholog of human MSH6, encodes the Msh2 partner that completes MutSalpha, contributing the conserved Phe-X-Glu mismatch-reading motif that stacks against the mispaired base and an amino-terminal PWWP domain that binds H3K36me3, thereby coupling mismatch repair to actively transcribed, replication-timed chromatin; it also participates in somatic hypermutation and in class switch recombination fidelity in B cells. Human MSH6 mutations produce an attenuated Lynch phenotype skewed toward endometrial cancer with weaker microsatellite instability, and mouse nulls are viable and tumor-prone with a correspondingly milder instability signature, because Msh3-containing MutSbeta still handles insertion and deletion loops. That partial coverage is a genuine redundancy caveat: Msh6 with Msh3 compound animals are needed to reproduce full MutS deficiency.
