Why model Nf2
Merlin, the product of mouse Nf2 and ortholog of human NF2, is a FERM domain cytoskeletal adaptor that couples cortical actin and adherens junctions to growth control, restraining the Hippo pathway by promoting LATS1 and LATS2 activation and YAP or TAZ phosphorylation, and separately entering the nucleus to inhibit the CRL4-DCAF1 E3 ligase. Biallelic inactivation underlies familial and sporadic schwannoma, meningioma, ependymoma, and a large fraction of malignant pleural mesothelioma, with tumorigenesis proceeding through unrestrained YAP-TEAD transcription rather than kinase hyperactivation. Germline nulls fail at gastrulation because of extraembryonic defects, so conditional nulls are mandatory: P0-cre or Postn-cre for Schwann cells, Wt1-cre for mesothelium, each yielding distinct tumor spectra. Compensation is limited, since the ezrin, radixin, and moesin family shares the FERM fold without recapitulating growth suppression. Choose nulls for tumor initiation and TEAD inhibitor sensitivity, and consider humanized alleles where clinical missense variants must be functionally classified.
