Nlgn3, the mouse ortholog of X-linked human NLGN3, encodes neuroligin-3, the only neuroligin found at both excitatory and inhibitory synapses, where its cholinesterase-like ectodomain binds neurexins and its cytoplasmic tail couples to PSD-95 or gephyrin scaffolds depending on synapse class. Neuroligin-3 is also required for tonic endocannabinoid signaling at hippocampal inhibitory synapses, and its ectodomain is cleaved by ADAM10 in an activity-dependent manner, with the shed fragment acting as a mitogen for glioma cells, a striking non-synaptic function. The autism-associated R451C substitution in the cholinesterase-like domain causes protein retention in the endoplasmic reticulum yet behaves as a gain of function in vivo, increasing inhibitory transmission and producing social behavior deficits, whereas the null shows a distinct and partly opposite profile, so knockin and deletion answer genuinely different questions. Because Nlgn1 and Nlgn2 occupy overlapping synapse classes, conditional alleles paired with cell-type Cre drivers and synapse-resolved recordings are the appropriate design.
