Why model Nlgn1

Nlgn1, the mouse ortholog of human NLGN1, encodes neuroligin-1, a postsynaptic cell adhesion molecule with an extracellular cholinesterase-like domain that binds presynaptic neurexins in a calcium-dependent manner and a short cytoplasmic tail whose PDZ-binding motif recruits PSD-95 and whose gephyrin-independent interactions organize the excitatory postsynaptic density. Neuroligin-1 is targeted specifically to glutamatergic synapses by inclusion of splice site B, which biases binding toward beta-neurexins lacking splice site 4, and it preferentially controls NMDA receptor content, so deletion reduces the NMDA to AMPA ratio and impairs long-term potentiation and spatial memory without changing synapse number substantially. The protein is also shed by matrix metalloproteinase 9 in an activity-dependent manner, a step required for plasticity. Conditional nulls with region-restricted Cre drivers assign these effects to defined circuits, while splice site or PDZ motif knockins separate adhesion from scaffolding. Nlgn2 and Nlgn3 occupy distinct synapse classes and only partially buffer loss, so measure receptor composition directly.
