TrkA, encoded by mouse Ntrk1 in orthology with human NTRK1, is a receptor tyrosine kinase with leucine-rich repeats and immunoglobulin-like C2 modules that binds nerve growth factor, dimerizes, and signals through SHC, FRS2, phospholipase C gamma, and the RAS-ERK and PI3K-AKT cascades to specify survival of nociceptive sensory and sympathetic neurons. Biallelic loss-of-function variants cause congenital insensitivity to pain with anhidrosis, whereas oncogenic activation occurs through in-frame fusions such as LMNA-NTRK1 and TPM3-NTRK1 that dimerize constitutively via partner coiled coils, defining a tumor-agnostic indication for larotrectinib and entrectinib along with solvent front resistance at G595. Design therefore separates three questions: nulls for sensory neuron development and pain phenotypes, inducible fusion knockins for oncogenic dependency and inhibitor pharmacology, and humanized kinase domains for potency and resistance profiling. Germline nulls survive to birth but die early with severe sensory ganglion loss, so tissue-restricted deletion is preferable, and Ntrk2 and Ntrk3 bind distinct neurotrophins without compensating.
