Optineurin, encoded by mouse Optn and orthologous to human OPTN, is a coiled-coil autophagy receptor carrying an LC3-interacting region, a UBAN motif that binds linear M1-linked polyubiquitin, and a C-terminal zinc finger. TBK1 phosphorylates optineurin at Ser177 and adjacent residues to enhance LC3 binding and ubiquitin affinity, driving mitophagy downstream of PINK1 and parkin, xenophagy of cytosolic bacteria, and clearance of aggregated protein, while the UBAN domain simultaneously restrains NF-kappaB signaling. Disease logic is allele-specific: E50K acts as a dominant gain of function in normal-tension glaucoma, whereas E478G and truncating alleles cause ALS by abolishing ubiquitin binding. Model designs therefore fork between conditional null, UBAN point knockins such as D477N, phosphomimetic or phosphodead Ser177 variants, and humanization. Because Sqstm1, Nbr1, Tax1bp1, and Calcoco2 provide overlapping receptor activity, expect buffered clearance phenotypes and pair conditional alleles with motor neuron or microglial Cre drivers.
