Pdha1, the mouse ortholog of human PDHA1, encodes the E1 alpha subunit of the pyruvate dehydrogenase complex, which pairs with E1 beta in an alpha2beta2 heterotetramer, binds thiamine pyrophosphate, and performs the decarboxylation of pyruvate that commits carbon to acetyl-CoA and the TCA cycle. Regulation runs through three serine residues on this subunit that are phosphorylated by the PDK kinases and reversed by PDP phosphatases, making the protein the direct node where nutrient state gates glycolytic carbon entry into oxidative metabolism. The gene is X-linked, and PDHA1 deficiency is the commonest cause of congenital lactic acidosis, hemizygous in males and mosaic in heterozygous females, an architecture that should shape design: conditional floxed alleles permit tissue-restricted loss without the confound of random X inactivation, while patient-matched knockins retain residual activity for dichloroacetate or ketogenic diet studies.
