Pfkl is the mouse ortholog of human PFKL and encodes the liver-type subunit of phosphofructokinase 1, the tetrameric enzyme that commits glucose to glycolysis by phosphorylating fructose 6-phosphate to fructose 1,6-bisphosphate, the step under the strongest allosteric control by ATP, citrate, AMP and fructose 2,6-bisphosphate. The L subunit assembles into homotetramers and into hybrid tetramers with the M and P subunits, and the resulting isozyme composition sets tissue-specific sensitivity to those effectors, with the liver-enriched form tuned for gluconeogenic and lipogenic crosstalk. Because the human gene sits on chromosome 21, dosage effects have also been examined in trisomy contexts. Conditional nulls localize where L subunit content limits glycolytic flux, while humanization supports allosteric modulator screening. Since Pfkm and Pfkp form functional tetramers in overlapping tissues, single-locus deletion is frequently buffered and compound conditional alleles are advisable.
