Encoding urokinase-type plasminogen activator, Plau is the mouse ortholog of human PLAU and produces a single-chain zymogen that binds its GPI-anchored receptor uPAR (Plaur) through an N-terminal growth factor domain, is activated by plasmin or cathepsin cleavage, and then generates plasmin at the cell surface rather than in the fluid phase. This pericellular proteolysis drives matrix degradation, latent TGF-beta and pro-MMP activation, macrophage and smooth muscle migration, and integrin plus vitronectin dependent signaling that persists even when catalysis is blocked. Consequently a conditional null tests requirement in myeloid, endothelial, or tumor compartments, an active-site knockin separates proteolysis from uPAR occupancy, and humanization of the growth factor domain is necessary for antibodies or ATF fusions that ignore mouse protein. Plat and plasma kallikrein compensate in the fluid phase, so pair Plau alleles with invasion, fibrosis, or fibrinolysis endpoints and a validated lineage Cre.
