Why model Pml
Pml encodes the mouse ortholog of human PML (TRIM19), a RBCC tripartite motif protein whose coiled coil and SUMO to SIM interactions polymerize into PML nuclear bodies that concentrate p53, CBP, HIPK2, DAXX, and SP100 to coordinate apoptosis, senescence, antiviral responses, and stem cell quiescence. Its cardinal oncogenic role is as the 5 prime partner in t(15;17) PML-RARA, the driver of acute promyelocytic leukemia, where the fusion oligomerizes through the PML coiled coil, converts retinoic acid receptor alpha into a constitutive repressor of myeloid differentiation genes, and is uniquely susceptible to all-trans retinoic acid and to arsenic trioxide, which binds PML cysteines and triggers SUMO-dependent RNF4 degradation. Germline nulls are viable but tumor prone with impaired senescence, so conditional nulls address tumor suppressor and stem cell questions, whereas cathepsin G promoter driven or inducible PML-RARA knockins generate authentic promyelocytic leukemia. Because arsenic targeting depends on precise cysteine and SUMO site context, humanized PML alleles suit pharmacology endpoints.
