Why model Prr5
Protor-1, the product of Prr5 and ortholog of human PRR5, is an accessory subunit of mTOR complex 2 that binds the complex through RICTOR rather than mTOR itself and is dispensable for complex assembly, distinguishing it from RICTOR and mSIN1, whose loss dissolves mTORC2 entirely. Its documented function is substrate-selective: Protor-1 deficiency reduces mTORC2-dependent hydrophobic motif phosphorylation and activation of SGK1 in kidney while leaving AKT Ser473 phosphorylation largely intact, making the locus a rare tool for dissociating branches of mTORC2 output. Null animals are viable and grossly normal, so constitutive deletion is acceptable and conditional alleles are needed only for lineage-specific renal or metabolic questions. The paralog Prr5l encodes Protor-2 and occupies the same RICTOR interface, so compound deletion is advisable when total SGK1 signaling is the endpoint; match allele choice to whether the question concerns complex integrity or substrate steering.
