Why model Psen2
Second catalytic subunit of gamma-secretase, Psen2 is the mouse ortholog of human PSEN2 and assembles with nicastrin, APH1, and PEN2 into complexes localizing preferentially to late endosomes and lysosomes rather than the plasma membrane, giving PSEN2-containing gamma-secretase a distinct intracellular substrate pool and a documented role in mitochondria-associated ER membrane calcium handling. Rare dominant PSEN2 mutations, including N141I, cause early onset Alzheimer disease by shifting cleavage toward longer amyloid beta species. Unlike Psen1, Psen2 knockout mice are viable and fertile with only mild age-dependent pulmonary fibrosis, which makes constitutive null tractable, whereas Psen1 and Psen2 double nulls phenocopy severe Notch deficiency and die embryonically. Genuine redundancy with Psen1 therefore dictates a compound conditional strategy whenever total gamma-secretase output is the endpoint. Use patient knockins for amyloid ratio questions, conditional double alleles for substrate accumulation studies, and humanization when modulator pharmacology is the goal.
