Why model Relb

Non-canonical NF-kappaB signaling runs through Relb, the mouse ortholog of RELB, whose product pairs almost exclusively with p52 generated by NIK and IKK alpha dependent processing of the NFKB2 p100 precursor, a slow, protein-synthesis-dependent arm activated by lymphotoxin beta receptor, CD40, BAFF receptor and RANK rather than by TNF. RELB and p52 dimers drive lymphoid organogenesis, follicular and conventional dendritic cell maturation, and thymic medullary epithelial development including Aire expression, and RELB also forms inhibited complexes with p100 that sequester it until processing occurs. Constitutive nulls are viable but develop multiorgan inflammation, splenomegaly and disorganized secondary lymphoid architecture, so conditional alleles are chiefly useful for separating stromal from hematopoietic contributions. Because p100 acts as an inhibitor as well as a precursor, pair Relb alleles with Nfkb2 processing-dead knockins when the endpoint is pathway logic rather than simple loss of the transactivator.
