Ripk1 is the mouse ortholog of human RIPK1 and encodes a kinase whose N-terminal catalytic domain, ubiquitin-modified intermediate region, RHIM, and C-terminal death domain let it act as both scaffold and executioner downstream of TNFR1, TLR3, TLR4, and ZBP1. Within complex I, cIAP and LUBAC dependent ubiquitination of RIPK1 supports NF-kB and MAPK output, whereas loss of that modification allows cytosolic complex II formation, where autophosphorylated RIPK1 engages RIPK3 for necroptosis or FADD and caspase-8 for apoptosis, with caspase-8 cleavage at Asp325 serving as a brake. Nulls die perinatally from systemic inflammation, so the informative alleles are viable kinase-dead D138N knockins that isolate scaffold from catalytic function, RHIM mutants, cleavage-resistant D325A alleles, and tissue-restricted conditional deletion.
