Serving as the mouse ortholog of human RIPK3, Ripk3 encodes the obligate necroptosis kinase that oligomerizes with RIPK1, ZBP1, or TRIF through RHIM-mediated amyloid-like interactions and then phosphorylates MLKL at Thr231 and Ser345, triggering conformational change, membrane targeting, and lytic death with DAMP release. Kinase activity also supports inflammasome activation and cytokine transcription in some myeloid settings, so the protein is not purely a death switch. Genetically, nulls are viable and fertile and give clean epistasis, but kinase-dead D161N knockins are embryonic lethal because the inactive protein still assembles with RIPK1 and drives caspase-8 apoptosis, a contrast that is itself informative. Because caspase-8 restrains this axis, compound Casp8 or Mlkl alleles are frequently required for interpretation, and conditional deletion assigns requirement to intestinal epithelium, keratinocytes, or macrophages. Align allele class with ischemic injury, viral clearance, or MLKL phosphorylation endpoints.
