S100a8, the mouse ortholog of human S100A8, encodes MRP8 (calgranulin A), a small EF-hand protein that heterodimerizes obligately with S100A9 to form calprotectin, the dominant cytosolic cargo of neutrophils and inflammatory monocytes. Released calprotectin chelates zinc and manganese to starve bacteria and fungi, engages TLR4 and RAGE to amplify NF-kB driven cytokine output and endothelial activation, and promotes myeloid-derived suppressor cell accumulation in tumors, while intracellularly the complex supports NADPH oxidase assembly and tubulin dynamics. Notably, germline S100a8 disruption has been reported as early embryonic lethal with resorption, whereas S100a9 nulls are viable and lack detectable S100A8 protein because the partner is unstable alone, so conditional or myeloid-restricted deletion is the safer design.
