Sdha is the mouse ortholog of human SDHA and encodes the flavoprotein catalytic subunit of succinate dehydrogenase, the only enzyme shared by the TCA cycle and the electron transport chain, which oxidizes succinate to fumarate using a covalently bound FAD attached through a histidyl linkage installed by the SDHAF2 flavinylation factor. Electrons pass from FAD into the iron-sulfur relay of SDHB and onward to ubiquinone at the membrane anchor formed by SDHC and SDHD. Biallelic hypomorphic alleles cause Leigh syndrome and cardiomyopathy, whereas heterozygous germline loss with a somatic second hit predisposes to paraganglioma, gastrointestinal stromal tumor and renal tumors through succinate accumulation that inhibits alpha-ketoglutarate-dependent dioxygenases. Design accordingly: patient-matched knockins model the recessive bioenergetic disease, while conditional nulls in chromaffin or neural crest lineages model the tumor suppressor arm.
