Co-transcriptional R-loop resolution requires Setx, the mouse ortholog of human SETX, encoding senataxin, a large superfamily 1 helicase with a C-terminal DNA and RNA helicase domain and an N-terminal protein interaction region contacting RNA polymerase II, the RNA exosome, and DNA damage response factors. Senataxin unwinds RNA:DNA hybrids at transcription termination sites and at collisions between transcription and replication, limiting genome instability and supporting antisense transcript degradation and meiotic sex chromosome inactivation. Disease logic splits cleanly: recessive loss of function causes ataxia with oculomotor apraxia type 2, while dominant L389S and R2136H alleles cause juvenile ALS4 through an apparent gain or change of function. Allele design must therefore separate conditional null from dominant point knockins, since these model different diseases. Note that Setx null males are sterile with meiotic arrest, so conditional deletion with a neuronal Cre is preferable when motor neuron or cerebellar endpoints are the focus.
