VGLUT1 defines telencephalic excitatory identity and is developmentally exchanged for VGLUT2 at many synapses. Homozygous nulls survive birth but decline sharply after the third postnatal week with blindness, ataxia, and death, reflecting that developmental switch, so conditional alleles are strongly preferred for adult circuit work. Knockin Cre or reporter alleles at the locus provide clean access to cortical excitatory populations. Because VGLUT2 partially substitutes at a subset of synapses during early development, interpret quantal size and vesicle filling phenotypes with that transient overlap in mind and select region-specific Cre drivers accordingly.
