Serotonin reuptake is executed by Slc6a4, the mouse ortholog of human SLC6A4, encoding SERT, an SLC6 family sodium and chloride dependent symporter with a twelve-transmembrane LeuT fold, a central substrate site, and an allosteric vestibular site, expressed on raphe serotonergic terminals as well as platelets, gut, and transiently on developing thalamocortical and sensory neurons. SERT is the direct target of SSRIs, MDMA, and cocaine, and it is regulated by PKC-dependent internalization, p38 MAPK phosphorylation, and syntaxin 1A interaction. Human variation includes the 5-HTTLPR promoter polymorphism and rare coding variants such as I425V. Slc6a4 knockout mice are viable with elevated extracellular serotonin, blunted 5-HT1A autoreceptor function, anxiety-like behavior, and abnormal barrel cortex patterning, making the null informative but developmentally confounded. Conditional alleles with tamoxifen-inducible Pet1-CreERT2 separate adult from developmental roles, and humanized alleles are preferred for SSRI pharmacology and promoter variant modeling.
