Srebf1, ortholog of human SREBF1, encodes membrane-bound basic helix-loop-helix leucine zipper transcription factors, SREBP-1a and the liver and adipose enriched SREBP-1c, produced from alternative promoters and first exons. Both are synthesized as endoplasmic reticulum precursors held by SCAP and, when sterols are abundant, retained through INSIG binding; sterol depletion permits COPII transport to the Golgi, sequential cleavage by S1P and S2P, and release of the nuclear form that occupies sterol response elements to drive Acaca, Fasn, Scd1 and Elovl6. SREBP-1c is additionally induced by insulin through AKT and by LXR agonism, making the locus the transcriptional hub of de novo lipogenesis. Hepatocyte or adipocyte conditional nulls address tissue-autonomous lipogenesis, while nuclear-form transgenes test sufficiency without processing control. Srebf2 partially substitutes at shared promoters, so measure both nuclear forms before attributing altered fatty acid synthesis to Srebf1 alone.
