Srsf2 encodes the mouse ortholog of human SRSF2 (SC35), a serine and arginine rich splicing factor with a single RNA recognition motif and a phosphorylated RS domain that binds exonic splice enhancers, recruits U1 and U2 snRNP to nascent transcripts, and also couples to transcriptional elongation and R loop resolution. The recurrent P95H, P95L, and P95R substitutions in myelodysplastic syndrome, chronic myelomonocytic leukemia, and secondary acute myeloid leukemia are heterozygous and mutually exclusive with SF3B1 and U2AF1 lesions, acting by shifting exonic enhancer preference from GGNG to CCNG motifs, which drives poison exon inclusion in Ezh2 and aberrant splicing of Bcor rather than simple loss of activity. Model design therefore requires conditional heterozygous P95H knockins, since homozygous mutant and null alleles are not tolerated in hematopoietic cells, with Mx1-cre or Vav1-cre induction to capture dysplasia and clonal advantage. Align allele choice with endpoint, whether splicing catalogs, competitive transplantation, or PRMT5 and spliceosome inhibitor sensitivity.
