Tet2 is the mouse ortholog of human TET2 and encodes an iron and 2-oxoglutarate dependent dioxygenase that oxidizes 5-methylcytosine to 5-hydroxymethylcytosine and further products, initiating active and passive demethylation at enhancers; unlike Tet1 and Tet3 it lacks a CXXC domain and instead relies on partners such as IDAX, WT1, and EBF1 for locus targeting. Truncating and catalytic domain missense mutations make TET2 one of the most frequent drivers of clonal hematopoiesis and of myelodysplastic syndrome, chronic myelomonocytic leukemia, and angioimmunoblastic T cell lymphoma, and its activity is phenocopied by IDH1 or IDH2 neomorphic mutations through 2-hydroxyglutarate inhibition while being stimulated by ascorbate. Germline nulls are viable with progressive hematopoietic stem cell expansion, myeloid bias, and long tumor latency, so conditional nulls with Vav1-cre or Mx1-cre plus a cooperating Flt3 internal tandem duplication or Jak2 V617F allele sharpen the phenotype. Paralogs Tet1 and Tet3 partially buffer hydroxymethylation, making compound conditional alleles worthwhile.
