The classic Lps2 frameshift allele established that TRIF loss abolishes poly(I:C) responses and the late-phase LPS interferon response while sparing early MyD88 signaling. Nulls are viable, so germline deletion is tractable, whereas RHIM or TRAF-binding knockins separate interferon induction from death signaling and Myd88 compound alleles are needed because both adaptors serve TLR4.
