Xiap is the mouse ortholog of human XIAP and encodes an inhibitor of apoptosis protein with three BIR domains, a ubiquitin-associated region, and a C-terminal RING E3 ligase, in which BIR2 plus its preceding linker occludes the active sites of caspase-3 and caspase-7 while BIR3 binds and holds caspase-9 monomers inactive, and the RING ubiquitinates caspases and other substrates for degradation. SMAC (Diablo) released from mitochondria displaces these interactions through its IAP-binding motif, the principle exploited by SMAC mimetics. Independently of apoptosis, XIAP ubiquitinates RIPK2 to sustain NOD1 and NOD2 driven NF-kB signaling, which is why human loss of function causes X-linked lymphoproliferative syndrome type 2 with hemophagocytic lymphohistiocytosis and Crohn-like disease.
