Zbtb16 is the mouse ortholog of human ZBTB16 (PLZF), a transcriptional repressor combining an N-terminal BTB or POZ dimerization domain with nine C-terminal C2H2 zinc fingers, which recruits NCOR1, SMRT, HDAC, and PRC1 components to target promoters and enforces spermatogonial stem cell self-renewal, innate effector programming of invariant natural killer T cells, and limb and axial skeletal patterning. In oncology it appears as the 5 prime partner of the t(11;17) ZBTB16-RARA fusion in a rare, all-trans retinoic acid resistant variant of acute promyelocytic leukemia, where the BTB domain drives constitutive oligomerization and corepressor recruitment that pharmacologic retinoid concentrations cannot dislodge. Germline nulls are viable, phenocopying the luxoid mutant with limb and vertebral defects and progressive loss of spermatogonial stem cells, so conditional nulls suit adult germline and immune endpoints, whereas inducible fusion knockins address differentiation therapy resistance. No close paralog substitutes for this BTB and zinc finger combination, so single-locus deletion is interpretable with a lineage-matched Cre.
