Why model Acvr2a
Encoding the activin type IIA receptor, Acvr2a is the mouse ortholog of human ACVR2A and produces a constitutively active serine/threonine kinase that captures activin A and activin B, plus myostatin, GDF11 and several BMPs, then transphosphorylates the GS box of ALK4, ALK5 or ALK7 to launch SMAD2 and SMAD3 signaling. Ligand affinity is tuned by follistatin sequestration and by the accessory coreceptor cripto, so ligand trap experiments and receptor genetics report different layers of the same axis. Conditional null alleles resolve tissue-autonomous requirements in gonadotropes, where loss blunts FSH synthesis, and in craniofacial mesenchyme, whereas extracellular domain humanization supports pharmacokinetic testing of receptor Fc decoys that are notoriously cross-reactive. Because Acvr2b binds an overlapping ligand set and largely compensates in muscle and skeleton, single knockouts underreport phenotype severity.
