As the dominant AGC kinase isoform in most tissues, Akt1 is the mouse ortholog of human AKT1 and encodes a PH domain kinase recruited to PIP3 generated by class I PI3K, activated by PDK1 phosphorylation of T308 in the activation loop and mTORC2 phosphorylation of S473 in the hydrophobic motif. Active AKT1 phosphorylates TSC2, PRAS40, GSK3 beta, BAD, and FOXO factors, coupling growth factor input to mTORC1 output, glycogen metabolism and apoptotic threshold. The recurrent E17K substitution in the PH domain locks the kinase at the membrane and drives Proteus syndrome and multiple carcinomas, so a conditional knockin of that codon addresses hyperactivation while a floxed null addresses requirement. Homozygous null mice are viable but growth restricted, and Akt2 and Akt3 compensate in muscle, liver and brain respectively, so isoform-specific conclusions demand compound alleles. Pair allele class to readout: null for dependency, E17K knockin for inhibitor pharmacodynamics.
