Why model Arid1a
Arid1a encodes BAF250a, the mouse ortholog of human ARID1A and the largest accessory subunit of the canonical BAF chromatin remodeling complex, where its ARID domain contributes non-sequence-specific DNA contact and its C-terminal region docks onto the SMARCA4 ATPase to target enhancer nucleosome eviction. Tumor alleles are overwhelmingly nonsense or frameshift, collapsing accessibility at lineage and differentiation enhancers in ovarian clear cell carcinoma, endometrioid tumors, gastric adenocarcinoma, and hepatocellular carcinoma. Homozygous deletion causes gastrulation-stage embryonic lethality near E6.5, so useful models require conditional flox alleles with tissue-restricted Cre, frequently combined with Pten or Trp53 loss for penetrant tumors.
