Why model C3
C3, ortholog of human C3, encodes the pivotal complement component whose proteolytic activation by either the classical and lectin pathway C4b2a convertase or the alternative pathway C3bBb releases the C3a anaphylatoxin and exposes a reactive thioester in the C3b thioester-containing domain that covalently tags surfaces. Deposited C3b seeds further convertase assembly for amplification, is degraded by factor I to iC3b and C3dg for CR3, CR4, and CR2 dependent phagocytosis and B cell priming, and feeds C5 convertase formation. Loss produces pyogenic infection susceptibility, impaired immune complex clearance, and blunted antibody responses, while dysregulated turnover underlies C3 glomerulopathy and paroxysmal nocturnal hemoglobinuria pathology. No paralog compensates, making the null definitive. Humanization is the design of record for compstatin family peptides and pegcetacoplan, which bind human C3 and do not engage the mouse protein, whereas hepatocyte-conditional deletion resolves liver-derived versus local myeloid C3 contributions.
