Three distinct human mutation classes map to different alleles: truncating loss of function causes episodic ataxia type 2, gain-of-function substitutions such as R192Q and S218L cause familial hemiplegic migraine type 1, and a C-terminal polyglutamine expansion causes spinocerebellar ataxia type 6. Constitutive nulls develop severe ataxia and dystonia and die around weaning, so cerebellar conditional deletion using Pcp2-Cre or granule cell drivers is required for adult physiology. CaV2.2 partially substitutes at some central terminals, so record paired-pulse and calcium imaging readouts rather than relying on transmitter release amplitude alone.
