Why model Cdk6
Cdk6 is the mouse ortholog of human CDK6 and encodes the second D-cyclin dependent kinase, sharing RB directed catalytic function with CDK4 and INK4 sensitivity but distinguished by dominant expression in hematopoietic stem and progenitor cells and lymphocytes, and by a kinase-independent nuclear function in which CDK6 acts as a transcriptional cofactor with STAT3, AP-1 and NF-kB to induce VEGFA and to repress p16 and p53 outputs. Chromosomal rearrangements placing CDK6 under strong enhancers occur in lymphoid malignancy, and CDK6 upregulation is a recurrent mechanism of acquired resistance to CDK4/6 inhibitors in breast cancer. Null mice are viable with mild anemia and thymic hypoplasia, so constitutive alleles are workable, but Cdk4 compensates in most nonhematopoietic tissue and single deletions mislead. Kinase-dead knockin alleles are the decisive tool here because they separate catalytic from transcriptional function, a distinction no null can make, while conditional nulls layered on Cdk4 alleles serve inhibitor resistance and hematopoietic dependency studies.
