Why model E2f2

Encoding a second activator E2F, E2f2 is the mouse ortholog of human E2F2 and produces a DP-heterodimerizing transcription factor with a winged helix DNA binding domain, a marked box domain mediating cofactor contacts, and a C-terminal transactivation and RB pocket binding region, occupying the same S phase promoters as E2F1 and E2F3 downstream of CDK dependent RB inactivation. Its distinguishing biology is immunological: E2F2 restrains T lymphocyte proliferation and null mice develop enhanced T cell activation with autoantibody production and a lupus-like autoimmune syndrome, and it also contributes to pancreatic beta cell mass. Because nulls are viable, constitutive alleles suffice for immunology work, while conditional alleles are preferable when separating lymphoid from stromal contributions. Redundancy within the activator subfamily is substantial, since E2f1 and E2f3 occupy overlapping targets, so proliferative phenotypes appear only in compound backgrounds.
