Rare human ENO1 loss causes hemolytic anemia, while ENO1 deletion in glioma creates collateral lethality with ENO2 dependence. Allele strategy divides sharply: a conditional null tests glycolytic dependence within a defined lineage, a catalytically dead knockin separates enzymatic from moonlighting functions, and humanization enables evaluation of enolase inhibitors with human-selective pharmacology. Because Eno2 and Eno3 substitute in neurons and muscle respectively, confine conclusions to tissues where alpha-enolase dominates, and prioritize flux measurements over transcript abundance as endpoints.
