Eno1, the mouse ortholog of human ENO1, encodes alpha-enolase, the magnesium-dependent dimeric lyase that dehydrates 2-phosphoglycerate to phosphoenolpyruvate in the payoff phase of glycolysis and supplies substrate to pyruvate kinase. Beyond catalysis the locus produces MBP-1 by alternative translation, a nuclear repressor acting at the MYC P2 promoter, and surface-displayed alpha-enolase serves as a plasminogen receptor on leukocytes and tumor cells, so phenotypes may be metabolic, transcriptional or proteolytic. Rare human ENO1 loss causes hemolytic anemia, while ENO1 deletion in glioma creates collateral lethality with ENO2 dependence. Allele strategy divides sharply: a conditional null tests glycolytic dependence within a defined lineage, a catalytically dead knockin separates enzymatic from moonlighting functions, and humanization enables evaluation of enolase inhibitors with human-selective pharmacology. Because Eno2 and Eno3 substitute in neurons and muscle respectively, confine conclusions to tissues where alpha-enolase dominates, and prioritize flux measurements over transcript abundance as endpoints.
