Why model F13a1
F13a1 is the mouse ortholog of human F13A1 and encodes the catalytic A subunit of factor XIII, a calcium-dependent transglutaminase circulating as an A2B2 heterotetramer and present intracellularly as cytoplasmic A2. Thrombin cleavage of the activation peptide plus calcium-driven dissociation from the B subunit exposes the cysteine active site, which forms gamma-glutamyl-epsilon-lysyl isopeptide crosslinks between fibrin gamma and alpha chains and incorporates alpha-2 antiplasmin, conferring mechanical strength and fibrinolysis resistance on the clot; additional substrates include factor V, von Willebrand factor, and matrix proteins, while macrophage and platelet A subunit contributes to wound healing and phagocytosis. Human A subunit deficiency causes severe bleeding with umbilical hemorrhage and poor healing, and mouse nulls are viable but bleed and show impaired uterine and cardiac repair. The B subunit cannot substitute catalytically. Use nulls or patient knockins for clot stability endpoints and conditional alleles to separate hepatic from myeloid A subunit pools.
