The carrier B subunit of factor XIII is encoded by mouse F13b, ortholog of human F13B, a plasma glycoprotein composed of ten sushi domains, also called complement control protein modules, that binds the catalytic A2 dimer to form the circulating A2B2 heterotetramer. The B subunit has no transglutaminase activity of its own; its role is to stabilize A2 in plasma, extend its half-life several fold, prevent premature calcium-dependent activation, and modulate presentation of factor XIII to fibrin, dissociating once thrombin cleaves the A subunit activation peptide so catalysis can proceed. Human F13B deficiency therefore produces a secondary reduction in circulating A subunit and a milder bleeding phenotype than A subunit deficiency, an informative dissociation between availability and catalysis. No paralog performs this carrier function.
