The same protein is the IGRP antigen recognized by pathogenic CD8 T cells in NOD mice, giving the locus a second, immunological identity. Consequently a straight null suits secretion threshold studies, humanized knockins are the right tool for validating human coding variants and epitope-matched T cell work, and conditional alleles with Ins1-Cre confirm beta cell autonomy.
