Why model Gabre

Gabre encodes the ε subunit, ortholog of human GABRE, an unusual GABA-A receptor subunit that substitutes for γ2 in pentamers expressed in locus coeruleus, dorsal raphe, hypothalamus and selected brainstem autonomic nuclei, situated in the X-linked cluster with Gabra3 and Gabrq. Its defining property is that ε-containing receptors display prominent spontaneous, agonist-independent channel opening and are largely insensitive to benzodiazepines and to the potentiating effects of anesthetics and neurosteroids, so they set a constitutive chloride leak in monoaminergic neurons rather than transducing phasic synaptic input. This makes them a plausible substrate for the differential anesthetic sensitivity of arousal circuits. Because loss removes a tonic conductance rather than a synaptic current, a conditional null crossed to Dbh-Cre or Slc6a4-Cre paired with whole-cell recording of holding current is the informative design, while humanization supports pharmacology on a subunit whose properties diverge across species. Verify ε expression by in situ before phenotyping.
