Because clinical antibodies do not cross-react, humanization is the allele for pharmacology, while conditional nulls with Cd4-Cre or Lyz2-Cre separate lymphoid from myeloid contributions, and combining with Pdcd1 loss is usually required before checkpoint phenotypes become measurable.
