The mouse Igf1r locus, ortholog of human IGF1R, specifies a disulfide-linked alpha2beta2 receptor tyrosine kinase that binds IGF-1 and IGF-2, autophosphorylates activation loop, juxtamembrane and C-terminal tyrosines, and recruits IRS1, IRS2 and SHC to drive PI3K to AKT and ERK output, and it also forms hybrid receptors with the insulin receptor that blur ligand specificity in tissues expressing both. Null mice are severely growth retarded and die neonatally of respiratory failure, so viable interrogation requires conditional alleles, with muscle, adipose, neuronal or endothelial Cre lines separating growth, metabolic and vascular contributions and inducible Cre avoiding developmental confounds. Kinase-dead and epitope-humanized knockins address different questions, the former testing scaffold versus catalytic function and the latter enabling preclinical evaluation of IGF1R-directed antibodies and inhibitors whose selectivity against the insulin receptor is the key liability. Because Insr forms hybrids and signals to the same effectors, plan compound Igf1r and Insr alleles before interpreting insulin sensitivity endpoints.
