Why model Igf2
Igf2 is the mouse ortholog of human IGF2 and encodes a fetal growth factor whose expression is paternally monoallelic under control of the H19 imprinting control region through CTCF-dependent enhancer competition, with additional input from intragenic differentially methylated regions. The mature peptide signals through IGF1R and, distinctively, through the insulin receptor A isoform, while IGF2R clears it from circulation, making its dosage unusually sensitive to both imprinting status and receptor stoichiometry. Human loss of imprinting drives Beckwith-Wiedemann overgrowth and Wilms tumor, whereas hypomethylation reduces growth in Silver-Russell syndrome, a bidirectional disease logic that allele design should mirror: a paternal-allele conditional null tests the growth requirement, imprinting control region deletion or targeted methylation alleles model biallelic expression, and humanization supports antibody or ligand trap testing.
