Why model Ikbkg
Encoding NEMO, the regulatory scaffold of the IkB kinase complex, Ikbkg is the mouse ortholog of human IKBKG and is X-linked; through its coiled-coil and leucine zipper UBAN region it reads Met1-linked linear polyubiquitin generated by LUBAC as well as Lys63 chains, and this ubiquitin recognition, rather than any catalytic function, is what juxtaposes and licenses IKK1 and IKK2 activation downstream of TNFR1, IL1R, Toll-like receptors, and antigen receptors. Human hypomorphic alleles cause anhidrotic ectodermal dysplasia with immunodeficiency, whereas amorphic alleles are male lethal and produce incontinentia pigmenti in heterozygous females, a pattern faithfully reproduced in mouse, where hemizygous nulls die in mid-gestation and heterozygous females show mosaic dermatopathy. Conditional deletion is therefore the workhorse design, UBAN point mutants such as the Asp311 substitution isolate ubiquitin sensing from scaffolding, and patient-matched hypomorphs model graded immunodeficiency. There is no paralog to buffer loss, so phenotypes report directly on allele strength and driver efficiency.
