Irf5 is the mouse ortholog of human IRF5 and encodes an interferon regulatory factor acting mainly downstream of MyD88 coupled TLR7 and TLR9 signaling, where TRAF6 dependent ubiquitination and IKKbeta mediated phosphorylation drive nuclear translocation and transcription of interleukin 6, interleukin 12, interleukin 23, and TNF, making it a determinant of inflammatory macrophage polarization and plasma cell differentiation rather than of type I interferon amplification. Human IRF5 haplotypes are among the strongest non-HLA risk factors for systemic lupus erythematosus, giving clear rationale for both loss-of-function and expression level alleles. One technical caveat dominates this locus: several widely used Irf5 knockout lines carried a closely linked spontaneous Dock2 mutation that independently distorts lymphocyte development and migration, so new projects should use a freshly targeted conditional allele and genotype Dock2 explicitly. Pair the flox with Lyz2-Cre or Cd19-Cre to separate myeloid from B lineage contributions, and humanize when antibody or degrader pharmacology is the endpoint.
