Why model Jag1
Jag1 is the mouse ortholog of human JAG1 and encodes a single-pass Notch ligand built from a DSL domain, sixteen EGF-like repeats, and a cysteine-rich region, which engages NOTCH1 through NOTCH4 in trans and requires mechanical force to expose the receptor S2 site for ADAM10, licensing gamma-secretase cleavage and NICD release, with Fringe glycosylation of the receptor tuning Jagged versus Delta-like preference and cis expression acting inhibitory. Heterozygous human loss of function causes Alagille syndrome with bile duct paucity, cardiac outflow defects, butterfly vertebrae, and posterior embryotoxon, while nulls die near E10.5 from failed vascular remodeling. Conditional deletion in portal mesenchyme, endothelium, or arterial mural cells is therefore mandatory, Alagille missense knockins distinguish trafficking-defective from binding-defective ligand, and humanization enables antibody and decoy testing. Jag2, Dll1, and Dll4 supply overlapping ligand activity, and sensitized Notch2 hypomorph backgrounds are often required to reveal biliary phenotypes.
