Trithorax-family biology begins with Kmt2a, the mouse ortholog of human KMT2A (MLL1), whose C-terminal SET domain deposits H3K4 methylation at Hox loci after taspase1 cleavage and reassembly of the N-terminal and C-terminal fragments, with catalysis requiring the WRAD module (WDR5, RBBP5, ASH2L, DPY30) and locus targeting through menin, LEDGF, and CXXC-mediated CpG binding. Leukemogenesis proceeds not by point mutation but by 11q23 translocation, generating in-frame fusions such as MLL-AF9 or MLL-AF4 that retain the CXXC domain, recruit DOT1L and the super elongation complex, and enforce HOXA9 and MEIS1 expression. Allele design splits accordingly: conditional nulls test methyltransferase dependence and hematopoietic stem cell output, SET-dead knockins isolate catalysis, and inducible fusion knockins at the endogenous locus produce latency-appropriate leukemia. Germline nulls are embryonic lethal near midgestation while heterozygotes show skeletal and hematopoietic haploinsufficiency, and the paralog Kmt2b partially overlaps, so compound conditionals with a validated hematopoietic Cre are advisable.
