Why model Proc

Encoding protein C, Proc is the mouse ortholog of human PROC and yields a vitamin K dependent zymogen with a Gla domain, two EGF modules, and a chymotrypsin-like protease domain that is activated when thrombin, presented by endothelial thrombomodulin and concentrated by EPCR, removes the activation peptide. Activated protein C then uses protein S as cofactor to proteolyze factors Va and VIIIa, shutting down thrombin amplification, and separately signals through EPCR-dependent PAR1 cleavage to stabilize endothelial barrier function and dampen NF-kB driven cytokine output. Complete deficiency in mice is perinatal lethal from consumptive coagulopathy, so severe models rely on hypomorphic alleles giving a few percent of normal activity, hepatocyte-restricted conditional deletion with Alb-Cre, or signaling-selective knockins that retain cytoprotection without anticoagulant function. Select by endpoint, whether microvascular thrombosis, sepsis survival, or permeability, since the anticoagulant and cytoprotective arms segregate genetically.
