S6 kinase 1, encoded by Rps6kb1 and orthologous to human RPS6KB1, is an AGC kinase recruited to mTORC1 through a TOS motif that binds RAPTOR, then activated by mTOR phosphorylation of the Thr389 hydrophobic motif followed by PDPK1 phosphorylation of Thr229 in the activation loop. Its substrates include ribosomal protein S6, eIF4B, PDCD4, eEF2 kinase and IRS-1, the last constituting the negative feedback loop that limits PI3K signaling and underlies paradoxical insulin sensitization by rapamycin. Nulls are viable but small with hypoinsulinemia and reduced beta cell size, so conditional alleles matter mainly for tissue-autonomous growth questions, while a Thr389 phospho-dead knockin isolates mTORC1 input from PDPK1 input in a way inhibitors cannot. Because Rps6kb2 phosphorylates overlapping substrates and is upregulated after Rps6kb1 loss, compound alleles or an Rps6 phospho-dead knockin should be planned when translation output is the endpoint.
