Rps6kb2 encodes S6 kinase 2, the ortholog of human RPS6KB2, an AGC kinase that shares the TOS motif, RAPTOR docking and mTORC1-dependent hydrophobic motif phosphorylation with S6K1 but carries a carboxy-terminal nuclear localization signal, lacks the amino-terminal extension that targets S6K1 to the nucleolus, and accounts for a substantial fraction of ribosomal protein S6 phosphorylation in many tissues. Single deletion produces essentially no growth phenotype and is accompanied by increased S6K1 activity, a compensatory relationship that is the central design consideration here, since combined Rps6kb1 and Rps6kb2 deletion causes perinatal lethality and abolishes S6 phosphorylation. Practical strategies therefore use conditional alleles at both loci with a common Cre driver, or an Rps6 serine to alanine knockin when the question concerns S6 phosphorylation rather than kinase identity.
